More information is needed. NR is similar to vitamin Niacin or Nicotinamide. Vitamins causing cancer is the implication so must see the entire study and doses used. Anyone have a link to the complete study??

I, too, was concerned about this article since I take regular nicotinamide almost every day so I kept looking and found this ..

"Nicotinamide, also known as niacinamide, is an amide of nicotinic acid, which is commonly known as niacin. Nicotinamide riboside (NR), on the other hand, is a chemically altered form of nicotinamide that has unique attributes.Nov 14, 2019"

This made me feel a little better but I am still interested in a link to the whole study.

When I gave my elderly mother the Nicotanimide Riboside the skin cancers on her arm “ took off”& were quite extensive when excised. Also
“ horny” type lesions came up on her arms. I stopped it immediately!
I do take Niacinamide now.

Thank you. Will do some research.

NR can increase NAD+ to NADH ratio - in every tissue and cell where it ends up - why is that so hard to understand?

Cancer cells can take advantage of that as well!

There is no escape from death by energy alone.

Summary: The commercial dietary supplement nicotinamide riboside, touted to improve cardiovascular and neurological health, may actually increase the risk of developing breast cancer that metastasizes to the brain.Popular Dietary Supplement Causes Cancer Risk and Brain Metastasis - Neuroscience News

I think this is part of the push against vitamin B3, as I mentioned in this thread below as well.

Can't get access to her the study yet, but I doubt NR actually causes cancer and metastases. If I have some free time left before the end of the year I may try a study with NR and niacinamide in the same TNBC mouse xenograft model they used and see what results I get.

It doesn’t cause cancer. But it can aggravate existing deranged cellular situations

I agree. Just for reference, here is a screenshot of my study with niacinamide + thiamine stopping the growth of human mantle cell lymphoma (xenograft model). Now, niacinamide is not NR as the former is an SIRT1 blocker (unmetabolized niacinamide, that is) while the latter is an activator, so this may turn out to be a crucial difference, We'll see, I will try comparing the two.

Could this have something to do with Ribose forming AGEs and feeding cancer cells?
An excerpt on D-Ribose:

CAUTION: Do not take D-ribose if you are younger than 60, and then only if monitored by a medical practitioner knowing expected length of life, and assessment of risk/benefit which includes analysis of overall lifetime dietary sugars, animal proteins and fats, and family history for cancers and dementia. These might indicate damage already present from AGEs. Wow, now that is a real bombshell.

Why? Research out of China reveals that D-ribose participates in forming AGEs faster than any other form of sugar. AGEs are indicated in many degenerative diseases, including Alzheimer's. ref Plus, cancer cells use more ribose than normal cells to grow faster. Thus, the under 60 should especially use with caution. Every little bit builds up and at some point, it may be too late to correct or mitigate these influences. ref

EVERYTHING IS ALWAYS A MATTER OF DEGREES. IF OVERALL SUGAR IN DIET IS LOW, RIBOSE USE WOULD HAVE LESS OF AN EFFECT. But, the typical American diet is fat too high in sugars and oxidizing stress that adding D-Ribose could push glycation activities into overdrive. If cancer not present, Ribose could help prevent, but if there is a family history, best to stay away.

Testing safety of Ribose might not have lasted long enough for AGEs to be detected, or even associated. Yes, most references used animal studies and more research in humans is vitally needed. Benefits have to be balanced against risks, and using age as a factor for how long it takes AGEs to form and Ribose to influence health. Or at what amount Ribose would impact AGEs formation compared to overall sugar amounts in diet. Short term use may be more appropriate. Here is the research for the DO NOT USE recommendation for RIBOSE. ref ref ref

One nutrient of value to help slow down or prevent AGEs formation, Carnosine. ref

Here is a study looking at the amount of AGEs already formed in different foods, often influenced by cooking methods and temperatures, and their effects on inflammatory markers after consumption by diabetics. ref Look at the changes on some of these inflammation markers. This is a very revealing important small study on the impact of diet and cooking methods on health. Foods with high AGEs already formed, increase the amount of AGEs the body produces.

Ribose and Cancer

It is of interest to note that ribose can play a positive role with potassium in cancer cells. At just the right concentration, ribose may carry potassium into cancer cells which slows the division rate of these cells. D-ribose also participates in Krebs pathway to supply energy to healthy cells, but cancer cells must hinder this uptake by healthy cells and take more for themselves. More to discover here.

So D-Ribose is something one should be cautious about. I wonder what Ray Peat's comment on potassium ascorbate and D-Ribose would be.

Chris Masterjohn’s take…

The Bottom Line

This new study raises the concern that as little as 23-29 milligrams of nicotinamide riboside per day could possibly fuel cancer growth and metastasis in women with triple-negative breast cancer.

More broadly, it raises the possibility that other forms of breast cancer might be fueled with nicotinic acid (often labeled “niacin”) or high protein intakes.

However, this is a mouse model and it cannot be used to confirm anything about what happens in humans.

More importantly, we have strong reasons to believe that what prevents cancer is radically different from what treats cancer. Many things that may fuel the growth of an established cancer are our best strategies to prevent a cancer from ever becoming established.

This study should not be used to suggest healthy people may develop cancer as a result of supplementing with nicotinamide riboside. In fact, we should be very interested in the possibility that nicotinamide riboside could help prevent cancer. Nicotinamide (not riboside) has actually been shown to help reduce the risk of melanoma in high-risk subjects.

I believe that women diagnosed with triple-negative breast cancer should avoid supplementing with nicotinamide riboside unless they have a very strong reason to do so, and should be aware that it might fuel cancer growth or metastasis. This is a low-confidence judgment. Future studies in humans could be used to confirm, refute, or revise this. Until such studies exist, future animal studies may contradict this one and cause me to put this judgment on hold.

More broadly, I believe we should investigate the potential for fasting and restriction of growth-promoting nutrients, as well as alternation between cancer-killing therapies and nourishment regimes, to help with established cancer.

We should all have some form of fasting-feeding cycle, but those of us without diagnosed cancer should focus primarily on meeting our nutrient needs and should not worry about this nicotinamide riboside study.
